چکیده

The hydrolysis studies reported in the preceding paper (Hawthorne, 1960) suggest that in liver phosphatidylinositol the phosphate group is linked to one of the cis-hydroxyl groups of the inositol, i.e. to positions 1(3)or 2-. Until recently, the inositol monophosphate isolated from phosphoinositides was believed to be optically inactive (Iselin, 1949; Hawthorne, 1955) and so liver phosphatidylinositol was thought to have the 2-structure (Hawthorne & Hubscher, 1959 a). It can be seen from the structure (I) that only the 2or 5-phosphate has

Enhanced phosphodiesteratic breakdown and turnover of phosphoinositides during reperfusion of ischemic rat heart.

In this study, we examined phosphoinositide metabolism during ischemia and reperfusion using an isolated and perfused rat heart. When myocardial phosphoinositides were prelabeled with [3H]inositol, reperfusion after 30 minutes of normothermic global ischemia resulted in significant accumulations of radiolabeled inositol phosphate, inositol bisphosphate, and inositol trisphosphate. Isotopic inco...

Phosphoinositides in the Flight Muscle of Periplaneta americana: Incorporation of Labelled Phosphate and Inositol

The content of phosphatidylinositol phosphate (DPI) in the flight musculature of the cockroach Periplaneta americana was found to be 30 nmol g ' wet weight, that of phosphatidylinositol-bis-phosphate (TPI) 6.5 nmol g wet weight. After administration of P-orthophosphate in vivo, the specific radioactiv ity (S.A.) of arginine phosphate reaches a plateau after 2 hours and remains constant for the...

Changes in the levels of inositol phosphates after agonist-dependent hydrolysis of membrane phosphoinositides.

The formation of inositol phosphates in response to agonists was studied in brain slices, parotid gland fragments and in the insect salivary gland. The tissues were first incubated with [3H]inositol, which was incorporated into the phosphoinositides. All the tissues were found to contain glycerophosphoinositol, inositol 1-phosphate, inositol 1,4-bisphosphate and inositol 1,4,5-trisphosphate, wh...

Gonadotropin releasing hormone stimulates the formation of inositol phosphates in rat anterior pituitary tissue.

The production of inositol phosphates in response to gonadotropin releasing hormone (GnRH) was studied in rat anterior pituitary tissue preincubated with [3H]inositol. Prelabelled paired hemipituitaries from prepubertal female rats were incubated in the presence or absence of GnRH in medium containing 10 mM-Li+ X Li+, which inhibits myo-inositol-1-phosphatase, greatly amplified the stimulation ...

Sustained elevation in inositol 1,4,5-trisphosphate results in inhibition of phosphatidylinositol transfer protein activity and chronic depletion of the agonist-sensitive phosphoinositide pool.

The 43 kDa inositol polyphosphate 5-phosphatase (5-phosphatase) hydrolyses the signalling molecules inositol 1,4,5-trisphosphate (Ins(1,4,5)P(3)) and inositol 1,3,4,5-tetrakisphosphate (Ins(1,3,4, 5)P(4)) in a signal-terminating reaction. We have utilised cell lines that stably underexpress the 43 kDa 5-phosphatase, as a model system to investigate whether Ins(1,4,5)P(3) can control the rate of...
